基于生物碱指纹图谱和多组分定量分析的吴茱萸质量评价

目的 建立吴茱萸生物碱类成分HPLC指纹图谱,测定其中吴茱萸碱、吴茱萸次碱、吴茱萸新碱3个生物碱的含量,并结合聚类分析对不同产地吴茱萸进行质量评价.方法 采用HPLC法,以吴茱萸碱为参照,绘制11批吴茱萸的指纹图谱,并进行相似度评价、聚类分析;采用HPLC测定3个生物碱类成分的含量.结果 建立了吴茱萸HPLC指纹图谱,确定了 11个共有峰,指认出4个峰,与对照指纹图谱的相似度在0.898~0.996之间,不同产地的吴茱萸所含生物碱类成分种类和数量基本一致,3个生物碱类成分的含量在较大范围内波动较大.11批样品可聚为3类.结论 该方法简单、可靠、重复性良好,可有效、全面用于吴茱萸质量的评价.
OBJECTIVE To establish fingerprints of Evodiae Fructus by HPLC,determine the content of evodiamine,rutaecarpine,evocarpine three alkaloids,and the quality of Evodiae Fructus from different producing companies was evaluated by cluster analysis.METHODS To draw 11 batches Evodiae Fructus of fingerprints by HPLC with evodiamine as the reference,followed by similarity evaluation and cluster analysis;HPLC was used to determine the content of three alkaloid components.RESULTS The HPLC fingerprint of Fructus Evodiae was established with 11 common peaks identified and 4 peaks identified.The similarity with the control fingerprint ranged from 0.898~0.996.The types and quantities of alkaloids in Evodiae Fructus from different regions were basically the same,and the content of the three alkaloids fluctuated significantly within a large range.11 batches of samples can be grouped into 3 categories.CONCLUSION This method is simple,reliable,and has good repeatability,and can be effectively and comprehensively used for the quality evaluation of Evodiae Fructus.
