艾绒总黄酮提取工艺优化及其抗氧化活性研究

目的 确定最佳的超声波提取艾绒中总黄酮的方法以及所得提取液的抗氧化活性.方法 首先,通过单因素实验和正交实验优化艾绒中总黄酮的提取工艺;其次,通过所得提取液对DPPH·和·OH的清除实验评价其抗氧化能力.结果 超声波辅助提取艾绒中总黄酮的最佳条件为:料液比为1∶60(g∶mL)、提取温度为50℃、超声时间为60min、乙醇浓度为50%.在此工艺下,艾绒中总黄酮的平均提取率为4.30%,RSD为0.40%.抗氧化实验结果表明,艾绒总黄酮提取液对DPPH·和·OH的IC50(半清除浓度)值分别为90.39μg·mL-1和185.51 μg·mL-1,表明艾绒总黄酮提取液具有一定的抗氧化能力.结论 该提取工艺操作简便,重复性良好,所得的提取液具有抗氧化活性,为艾绒资源的开发和使用提供指导.
Study on the optimization of extraction process and antioxidant activity of total flavonoids from moxa floss
OBJECTIVE To determine the optimal process for ultrasonic extraction of total flavonoids from moxa floss and the antioxidant activity of the obtained extract.METHODS First of all,the extraction process of total flavonoids in moxa floss was optimized by single factor and orthogonal experiments.Secondly,the antioxidant ability of the obtained extract was evaluated through the clearance experiments of DPPH and OH.RESULTS The optimal conditions for ultrasound-assisted extraction of total flavonoids from moxa floss were as follows:the material-liquid ratio of 1∶60(g∶mL),the extraction temperature of 50C,the ultrasound time of 60min,and the ethanol concentration of 50%.Under this process,the average extraction rate of total flavonoids from moxa floss was 4.30%,and the RSD was 0.40%.Moreover,the antioxidant test showed that flavonoids extracted from moxa floss had a certain antioxidant activity on DPPH and OH,with the IC50(semi-clearing concentration)values were 90.39μg·mL-1 and 185.51 μg·mL-1,respectively.CONCLUSION This method is simple and reproducible,and the obtained extract has antioxidant activity.They provide guidance for the development and application of moxa floss.
