Process optimization of astaxanthin extraction by a complex enzyme from Haematococcus pluvialis
Using astaxanthin extraction rate as the evaluation index,astaxanthin was extracted by the composite enzyme method,and five factors,namely,composite enzyme ratio,enzyme concentration,pH,reaction temperature,and reaction time,were selected to optimize the key process parameters through one-way and orthogonal tests.The experiment results showed that when the enzyme ratio was cellulose:pectinase=1∶1,the optimal process parameters were:enzyme concentration 7000U·mL-1,enzymatic pH=4.0,enzymatic temperature 50℃,enzymatic time 1h,and its average extraction rate was about 68.89%.conclusion:The process of astaxanthin extraction by composite enzyme method in this paper is feasible and can improve its extraction rate significantly,which can be practically applied to the extraction process of astaxanthin in red algae.
