小黄紫堇总生物碱的提取分离、结构鉴定及抗氧化活性研究

摘要
小黄紫堇为罂粟科紫堇属植物,是常用的广谱高效药物.对蒙药材小黄紫堇中的总生物碱进行提取分离,鉴别其总生物碱中化学成分并研究其抗氧化活性.首先将小黄紫堇全草醇浸后经酸化有机溶剂萃取,得到氯仿部分和正丁醇部分.其次对氯仿部分进行分离纯化并采用高效液相色谱-质谱、核磁共振等进行了表征.最终采用1,1-二苯基-2-甲基肼(DPPH)法测定了小黄紫堇总生物碱与单体生物碱对自由基的清除能力.实验结果表明,蒙药材小黄紫堇总生物碱提取率为1.185％,在总生物碱中发现一种单体生物碱,并证实了该物质为紫堇嘧啶碱(Corlumidin).通过实验发现小黄紫堇的总生物碱与单体生物碱Cor-lumidine均对DPPH自由基具有较强的清除能力.采用的研究方法简单,结果准确,为小黄紫堇的提取分离及其抗氧化活性的研究提供了有效依据.
Abstract
Coryddis raddeana Regel belongs to papaveraceae family and are widely-used and effective Mongolian and Tibet medical plants.The total alkaloids extracted and isolated from the mongolian herb Corydalis raddeana Regel,and identified the chemical components in the total alkaloids and assayed their antioxidant activity.Firstly,the entire grass from the mongolian herb Corydalis raddeana Regel was soaked in methanol and adjusted the pH with hydrochloric acid,then extracted with organic solvents to acquire Chloroform extracts and n-butanol extracts.Chloroform extracts was consequently isolated by semi-prepared column and identified the structure of components by LC-MS and NMR.Finally,The antioxidant activity of the total alkaloids and the isolated alkaloid were assayed by 11-diphenyl-2-picrylhydrazyl(DPPH)method.Results were indicated that the extraction rate of the mongolian herb Corydalis raddeana Regel was up to 1.185％,and identified one alkaloid,corlumidin.The antioxidant test indicated the total alkaloid and corlumidin possessed stong ability to scavenge DPPH free radicals.The research methods used in this study were simple and the results were accurate,which would provide a valid basis for the extraction,isolation and its antioxidant test of the mongolian herb Corydalis raddeana Regel.
