摘要

为确定矮丛风毛菊中多糖提取和醇沉的最优工艺参数,以矮丛风毛菊中多糖得率为响应值,采用苯酚-硫酸法测定多糖含量,单因素实验确定提取因素水平,正交设计方法优选最佳提取工艺[1].最佳提取工艺为:提取所用液料比w(H2O)∶w(矮丛风毛菊)为20∶1,提取次数为3次,提取时间为3 h;最佳醇沉工艺为:体积分数为80%的乙醇,试样浓缩液与乙醇体积比1∶6,醇沉静置时间15 min;最佳检测波长为490 nm.测得样品的平均多糖得率为8.87%.相对标准偏差(RSD)<2%,优选出的提取和醇沉工艺参数稳定可靠,该法准确、重复性好,为矮丛风毛菊开发利用提供了参考数据.

Abstract

The phenol-sulfuric acid method was used to determine the polysaccharide content,with the yield of polysaccharide as the response value so as to obtain the optimal process of polysaccharides extraction and alcohol precipitation from Saussurea eopygmaea Hand-Mazz.In addition,the single-factor experiment was used to determine the extraction factor level,and the orthogonal design method was used to optimize the extraction process.The best conditions were as follows:w(H2O)∶w(Saussurea eopygmaea Hand-Mazz)was 20∶1,extract three time(three hours every time);The optimum alcohol precipitation conditions were as flollows:volume fraction 80%ethanol,1∶6 volume ratio of concentrated solution and ethanol,and 15 min of ethanol settling time,and the optimal detection wavelength was 490 nm.It was found that polysaccharide content was 8.87%on average with a relative standard deviation(RSD)below 2%.The optimal parameter for the extraction and alcohol sinking process is stable and reliable,and the quantitative analysis method is accurate and reproducible for the Saussurea eopygmaea Hand-Mazz which provides reference data for the development and utilization.
