Recent Advances in the Novel Synthesis and Biomedical Applications of Polypeptides
Polypeptides are synthetic polymers with peptide bonds on the main chains.Due to their inherent biocompatibility and biodegradability, they have shown broad applications in the field of biomedicine. However, the efficient, fast and easy synthesis of polypeptides remains a challenge. In recent years, with the rapid development of synthetic chemistry, breakthroughs have been made in the preparation of polypeptides based on ring-opening polymerization of α-amino acid N-carboxyl anhydride(NCA),and a large number of novel materials based on polypeptides have been synthesized. In this review, the mechanism of synthesis of NCA monomers and the latest improvements were introduced firstly. Then the novel initiating or catalytic systems for the rapid and efficient synthesis of polypeptides by NCA ring-opening polymerization, which have been newly developed in the past few years was discussed specially. Finally, the typical applications of polypeptides in biomedical fields, including antibacterial agents, drug delivery and tissue engineering are described in brief. The future challenges of polypeptides-based materials for biomedical applications are also prospected.
