利用商品化妊娠试纸条的适配体传感器检测卡那霉素的研究

利用商品化妊娠试纸结合CRISPR/Cas12a系统检测卡那霉素。构建了包载人绒毛膜促性腺激素(HCG)的三维DNA水凝胶,利用卡那霉素和适配体特异性结合释放cDNA以激活CRISPR/Cas12a,剪切DNA水凝胶,从而使水凝胶破裂并释放HCG;释放的HCG可以利用商品化妊娠试纸进行可视化检测,结合智能手机与Image J软件也可以实现定量检测。
Detection of Kanamycin by Aptasensor using Commercial Pregnancy Test Strip
Kanamycin was detected using commercial pregnancy test strips combined with CRISPR/Cas12a system.First,three-dimensional DNA hydrogels containing human chorionic gonadotropin(HCG)were constructed.Then,specific binding of kanamycin to the aptamer released cDNA,which led to the activation of Cas12a.The activated Cas12a-crRNA cleaved the DNA hydrogel,leading to the release of HCG.Finally,the released HCG could be visually detected using commercial pregnancy test strips,and quantitative detection could also be achieved by combining smart phones and Image J software.
