Effect of Qingfei Tongluo Prescription on Pulmonary Fibrosis Following Acute Lung Injury in Rats
Objective:To investigate the inhibitory effect of Qingfei Tongluo Prescription on early pulmonary fi-brosis in rats with acute severe lung injury induced by endotoxin.Methods:Rats were randomly divided into six groups:blank group,model group,dexamethasone group,and Qingfei Tongluo Prescription low,medium,and high dose groups(10,20,30 g·kg-1).Pathological changes in lung tissue were observed using hematoxylin-eosin staining,and the degree of pulmonary fibrosis was analyzed by Masson staining.Plasma levels of IL-1β and TNF-αwere detected by ELISA.Results:Plasma levels of IL-1β and TNF-α in the model group were significantly higher than those in the blank group(P<0.05).In the Qingfei Tongluo Prescription groups(10,20,30 g·kg-1),IL-1β and TNF-α levels were significantly lower than those in the model group(P<0.05).Although IL-1β and TNF-α levels in the Qingfei Tongluo Prescription groups were lower than those in the dexamethasone group,the difference was not statistically significant(P>0.05).Hematoxylin-eosin staining showed that lung tissue damage in the dexa-methasone group and Qingfei Tongluo Prescription groups was significantly reduced compared to the model group,with preserved alveolar cavities and septa,and less alveolar thickening and fusion.Masson staining revealed sub-stantial collagen fiber deposition(stained blue)in the lung tissue of the model group,indicating increased fibrosis.In contrast,minimal blue staining and the least fibrosis were observed in the dexamethasone group,with similar findings in the Qingfei Tongluo Prescription groups.Conclusion:Qingfei Tongluo Prescription can reduce plasma levels of inflammatory factors IL-1β and TNF-α and inhibit early pulmonary fibrosis following acute lung injury in rats.
