Study on extraction and purification technique of rutin from Sophora japonica L.using acetone as solvent
In this study,acetone was used as the organic solvent for the extraction and purification of rutin from Sophora japonica L.by heating reflux method.The process conditions such as water bath temperature,water bath time,acetone concentration and material-to-liquid ratio were optimized by single-factor test and four-factor three-level response surface test design.The purification of crude rutin was performed by two recrystallization precipitation methods containing alkali dissolution-acid precipitation and acetone volatilization and then the purified rutin samples were detected by high performance liquid chromatography and nuclear magnetic hydrogen spectrometry.The results show that the results of the response surface optimization test are highly significant,and the optimal conditions of the extraction process are as follows water bath time 29 min,acetone concentration 54%,material-to-liquid ratio 8.6 g/L and water bath temperature 70.4 ℃,and the extraction rate of rutin is(79.17 ±0.91)%.The NMR spectra of rutin sample and the standard product are basically the same,and the purity of rutin is(96±0.81)%and the yield of pure product is(71±2.49)%.
