三七不同部位及果梗中皂苷的研究

比较三七不同部位(剪口、须根、主根、茎叶、果梗和花)中皂苷组成的差异.以HPLC方法,测定了果梗中Rb3、Fp2、Fc和Rd四种成分的含量;判断了三七不同部位中皂苷类成分的异同.用Agilent HC-C18色谱柱(4.6×250 mm,5 μm)分离;乙腈-水为流动相,梯度洗脱,流速:1.0 mL/min;检测波长:203 nm.发现三七地下部位中含有较多的三醇型皂甙,地上部位含有较多的二醇型皂甙.人参皂苷Rd、Rb3和三七皂苷Fc、Fp2在 0.2~2.0 mg/mL范围内线性关系良好(R2>0.9993),果梗中Rb3、Fp2、Fc和Rd的含量分别为1.4%、0.68%、0.71%和0.31%.三七地上部位与地下部位的化学成分差异较大;果梗、茎叶和花中皂甙成分极其相似,可以初步认为果梗能够代替茎叶提取皂苷.首次对果梗中Rb3、Fp2和Rd的含量进行了测定,且本实验建立的HPLC方法,为果梗的质量控制提供了一种简单、快速和灵敏的手段.
Study on saponins in different parts and fruit pedicel of notoginseng
The similarities and differences of saponins composition in different parts of Panax notoginseng(Burk.)F.H.Chen(including rhizome,fibrous root,main root,stem and leaf,fruit pedicel and flower)were analyzed in this paper.The contents of Rb3,Fp2,Fc and Rd in the fruit pedicel were determined.HPLC method was used to judge the saponin differences in different parts of P.notoginseng,and to determine the contents of Rb3,Fp2,Fc and Rd in fruit pedicel.Agilent HC-C18 column(4.6×250 mm,5 μ m)was used with acetonitrile-water as mobile phase,gradient elution.The flow rate was 1.0 mL/min and detection wavelength was set at 203 nm.There were more triol saponins in underground parts and more diol saponins in aboveground parts of notoginseng.Linear relationship of ginsenoside Rd,Rb3 and notoginsenoside Fc,Fp2 were good in the range of 0.2-2.0 mg/mL(R2>0.9993).The content of Rb3,Fp2,Fc and Rd were 1.4%,0.68%,0.71%and 0.31%in fruit pedicel,respectively.The chemical compositions of aboveground and underground parts of P.notoginseng were quite different.Saponin compositions of fruit pedicel,flower,stem and leaf were very similar,so it can be preliminarily considered that fruit pedicel can replace stem and leaf as saponin source.Contents of Rd,Rb3 and Fp2 in fruit pedicel were determined for the first time and the HPLC method established in this study provided a simple,rapid and sensitive mean for quality control of fruit pedicel of notoginseng.
