N-对香豆酰酪胺顺反立体异构化研究

反式N-对香豆酰酪胺(N-p-trans-coumaroyltyramine,N-p-t-CT),可以从多种植物中提取分离得到,但分离程序繁杂,得率很低,成本极高.研究表明N-p-t-CT具有抗氧化、抗炎、抗病毒、抗癌等多种药理活性.N-p-t-CT经光照可生成其顺式异构体N-对香豆酰酪胺(N-p-cis-Coumaroyltyramine,N-p-c-CT),且这两种立体异构体表现出不同的生物活性.为进一步研究其生物活性和作用机制,本文建立了N-p-t-CT的简单快速HPLC定量分析方法,并同时考察了不同光源、溶剂、温度、浓度及溶液pH等条件对其异构化的影响.研究表明,N-p-t-CT在避光、浓度高、溶剂极性小、碱性条件下更加稳定.研究结果为其进一步深入研究及相关产品研发提供了一定的理论依据和技术参考.
Studies on the Cis-trans geometric isomerization of N-P-coumaroyltyramine
N-p-trans--coumaroyltyramine(N-p-t-CT)can be extracted and isolated from various plants with complex separation process,low yield,and extremely high cost.Previous studies have stated that N-p-t-CT presents various pharmacological activities,including antioxidant,anti-inflammatory,antiviral,and anticancer effects.N-p-t-CT can generate its cis-isomer,N-p-cis-coumaroyltyramine(N-p-c-CT)under illumination,both of which exhibit different biological activities.In order to investigate the biological activities and mechanisms of N-p-t-CT,this study established a simple and rapid HPLC method for the quantitative analysis of N-p-t-CT.Meanwhile,the effects of different light sources,solvents,temperatures,concentrations,and pH on the degree of isomerization of N-p-t-CT were investigated.The results showed that N-p-t-CT was relatively stable under dark,high concentration,low solvent polarity and alkaline conditions,which provides a theoretical basis for further study and applications development.
