以槲皮素为先导化合物的抗癌剂的合成研究进展

槲皮素是天然抗氧化剂,主要存在于蔬菜、水果等植物中,可调节众多与疾病进展有关的细胞内外信号通路,刺激免疫系统,清除自由基,调节酶活性,改变有丝分裂周期,抑制、逆转或延缓肿瘤的发生.然而,槲皮素的生物利用度低,这限制了其在临床上的应用.研究人员通过各种方法设计和合成新的槲皮素衍生物,以改善其不利因素,进而发挥其预防和治疗疾病的作用.目前,已经合成了许多性能优良、溶解性能好、生物利用度高、代谢稳定、毒副作用小、抗癌活性显著的槲皮素衍生物.综述了近年来以槲皮素为先导化合物的合成,重点讨论了具有抗癌活性的槲皮素衍生物,为槲皮素衍生物的进一步开发提供参考.
Research Progress in the Synthesis of Anticancer Agents with Quercetin as the Lead Compound
Quercetin is a natural antioxidant,mainly found in vegetables,fruits and other plants.It can regulate many intracellular signaling path-ways related to disease progression,stimulate the immune system,scavenge free radicals,adjust enzyme activity,change the mitotic cycle,inhibit and reverse or delay the occurrence of tumors.However,the low bioavailability of quercetin limits its clinical application.New quercetin derivatives are designed and synthesized through various methods to improve its adverse factors and thereby play a role in preventing and treating diseases.Many quercetin derivatives have been synthesized with excellent properties,good solubility,high bioavailability,stable metabolism,low toxic and side ef-fects,and significant anti-cancer activity.The synthesis of quercetin as the lead compound in recent years is reviewed,focusing on quercetin deriva-tives with anti-cancer activity,and a reference for the further development of quercetin derivatives is provided.
