Determination of Chlorophyll a Concentration in Seawater by High Performance Liquid Chromatography Using Internal Standard Method
Chlorophyll a concentration is an important index of seawater quality monitoring and early warning of Marine red tide.High per-formance liquid chromatography is widely used as one of the most accurate measurement methods of Chlorophyll a concentration.β-Apoc-arotenal-8'-carotenal was selected as internal standard to establish a method for the concentration analysis of Chlorophyll a in seawater based on high performance liquid chromatographic internal standard method.In the experimental detection range,the correlation coeffi-cient was 0.999 8,the detection limit was 0.05 μg/L,and the recovery was 97.0%~100%.The relative standard deviation was 0.38%~0.55%,and the precision was good.At the same time,the significant difference between the internal and external standard methods was e-valuated by using the laboratory culture of chlorella algal fluid and the field seawater samples,and the results showed that there was no significant difference between the two methods.The results of internal standard method and external standard method in 12 groups of field seawater samples showed good correlation,the correlation coefficient was 0.92,and the average relative error was 6.21%.HPLC internal standard method for the determination of Chlorophyll a concentration in seawater has good separation effect and high accuracy,and can carry out more accurate quality control for the measurement of Chlorophyll a concentration in laboratory.
