Preparation of a multifunctional seaweed polysaccharide-based composite film and its protective effects against UVB-induced photoaging
Long-term exposure to ultraviolet B(UVB)radiation causes photoaging of the skin.Seaweed-derived polysaccharides are a class of structurally complex,acidic,marine polysaccharides with various biological activities such as antioxidant,anti-inflammatory,and immunomodulatory effects.Their oligosaccharides have better bioac-tivity and application potential due to low molecular weight and good water solubility.In this paper,a multifunc-tional seaweed polysaccharide-based composite film,the FAC film,was prepared using sulfated galactofucan poly-saccharides(DF)and oligosaccharides sodium alginate(AOS)as active ingredients,and κ-carrageenan(κ-Car)as the matrix.Next,their protective effects against UVB-induced skin photoaging were investigated via physico-chemical property characterization and in vivo experiments.The results of the physicochemical measurements showed that the film had good hygroscopic and moisturizing properties and a slow-release DF effect in different pH environments.The film exhibited good viscoelasticity after soaking in deionized water for 3 mins.The results of the in vivo experiments revealed that treatment with FAC film enhanced the skin water percentage and reduced UVB-induced thickening of the epidermis and dermis of the skin;elevated skin catalase(CAT)and superoxide dismutase(SOD)concentrations;suppressed expression of the pro-inflammatory factors interleukin-6(IL-6)and tumor necrosis factor(TNF-α);increased levels of the collagen characteristic amino acid hydroxyproline(HyP);and effectively reduced the expression of the collagen degrading enzyme substrate,metalloproteinase-3(MMP-3).These results suggested that the FAC film has excellent physicochemical properties such as moisturization,slower release,and ease of application,as well as appreciable biological activities of skin barrier repair and antioxidant,anti-inflammatory,and collagen degradation inhibition effects.Thus,it could be utilized as an agent for the preven-tion and treatment of skin photoaging.
