IL-1β、IL-6与TNF-α在脊髓损伤急性期与亚急性期中的表达研究

目的 探讨脊髓损伤(spinal cord injury,SCI)在急性损伤期与亚急性损伤期中白介素-1β(IL-1β)、白介素-6(IL-6)和肿瘤坏死因子(TNF-α)表达变化及其意义,为临床治疗脊髓损伤提供理论依据.方法 将Wistar雄性大鼠随机分为Sham组和SCI组,参考国际公认的改良Allen's造模法制作T9~T10脊髓损伤大鼠模型,分别在造模后1 d、6 d进行BBB评分;ELISA检测血清中IL-1β、IL-6和TNF-α的水平,分析变化规律.通过qRT-PCR检测大鼠脊髓中IL-1β、IL-6、TNF-α的基因表达,利用Western Blot来检测大鼠脊髓中IL-1β、IL-6、TNF-α和NF-κB的蛋白表达.分别记录两组治疗前和治疗后1个月、2个月、3个月的恢复情况、疼痛程度、生活质量等,并进行评估分析.结果 各组大鼠BBB评分比较差异具有统计学意义(P<0.05),与假手术相比,SCI组在各时间点组织IL-1β、IL-6、TNF-α的蛋白表达水平与基因表达水平显著增加,差异有统计学意义(P<0.05);并且在SCI各时间点,NF-κB蛋白表达水平与假手术相比显著增加,差异有统计学意义(P<0.005).与SCI-1 d相比,SCI-6 d中IL-1β、IL-6和TNF-α在蛋白与基因表达水平上均有适度降低,差异有统计学意义(P<0.05).结论 SCI在急性期与亚急性期中的病理学改变可能涉及脊髓内神经系统的损伤以及NF-κB信号通路的活化.SCI的继发性损伤在急性损伤期就已经发生,一直延续到慢性损伤期,可以通过外源性补充神经元细胞为SCI的治疗提供新的方向.
EXPRESSION OF IL-1β,IL-6 AND TNF-α IN ACUTE AND SUBACUTE SPINAL CORD INJURY
Objective To investigate the expression of interleukin-1β(IL-1β),interleukin-6(IL-6)and tumor necrosis factor(TNF-α)in acute and subacute spinal cord injury(SCI)and its significance,and to provide theoretical basis for clinical treatment of SCI.Methods Wistar male rats were randomly divided into Sham group and SCI group,and T9-T10 spinal cord inju-ry rat models were constructed according to the internationally recognized improved Allen's modeling method.BBB scores were performed at 1d and 6d after modeling,respectively.The levels of IL-1β,IL-6 and TNF-α in serum were detected by ELISA.Gene expressions of IL-1β,IL-6 and TNF-α in the spinal cord of rats were detected by qRT-PCR,and protein expressions of IL-1β,IL-6,TNF-α and NF-κB in the spinal cord of rats were detected by Western blot.Results BBB scores of all groups were statistically significant(P<0.05).Compared with sham operation,protein expression levels and gene ex-pression levels of IL-1 β,IL-6 and TNF-α were significantly increased in SCI group at each time point,and the dif-ferences were statistically significant(P<0.05).The expression level of NF-κB protein was significantly increased compared with sham operation(P<0.001).Compared with SCI-ld,the expression levels of IL-1β,IL-6 and TNF-α in SCI-6d were moderately decreased,and the differences were statistically significant(P<0.05).Conclusions The pathological changes of SCI in acute and subacute stages may involve the injury of the spinal cord nervous system and the activation of NF-κB signaling pathway.The secondary injury of SCI has occurred in the acute injury stage and continues to the chronic injury stage,which can provide a new direction for the treatment of SCI through exogenous neu-ron supplement.
