EXPRESSION OF IL-1β,IL-6 AND TNF-α IN ACUTE AND SUBACUTE SPINAL CORD INJURY
Objective To investigate the expression of interleukin-1β(IL-1β),interleukin-6(IL-6)and tumor necrosis factor(TNF-α)in acute and subacute spinal cord injury(SCI)and its significance,and to provide theoretical basis for clinical treatment of SCI.Methods Wistar male rats were randomly divided into Sham group and SCI group,and T9-T10 spinal cord inju-ry rat models were constructed according to the internationally recognized improved Allen's modeling method.BBB scores were performed at 1d and 6d after modeling,respectively.The levels of IL-1β,IL-6 and TNF-α in serum were detected by ELISA.Gene expressions of IL-1β,IL-6 and TNF-α in the spinal cord of rats were detected by qRT-PCR,and protein expressions of IL-1β,IL-6,TNF-α and NF-κB in the spinal cord of rats were detected by Western blot.Results BBB scores of all groups were statistically significant(P<0.05).Compared with sham operation,protein expression levels and gene ex-pression levels of IL-1 β,IL-6 and TNF-α were significantly increased in SCI group at each time point,and the dif-ferences were statistically significant(P<0.05).The expression level of NF-κB protein was significantly increased compared with sham operation(P<0.001).Compared with SCI-ld,the expression levels of IL-1β,IL-6 and TNF-α in SCI-6d were moderately decreased,and the differences were statistically significant(P<0.05).Conclusions The pathological changes of SCI in acute and subacute stages may involve the injury of the spinal cord nervous system and the activation of NF-κB signaling pathway.The secondary injury of SCI has occurred in the acute injury stage and continues to the chronic injury stage,which can provide a new direction for the treatment of SCI through exogenous neu-ron supplement.
