Modified Xiaochaihu Decoction Combined with Spine Pinching Treats Myocarditis Based on the Theory of"Zang-Fu Extraordinary Connection"
This study aims to explore the mechanism of modified Xiaochaihu Decoction combined with spine pinching in the treatment of myocarditis(MC).Thirty mice were randomized into blank, model,Western medicine, modified Xiaochaihu Decoction, spine pinching, and combined treatment(modified Xiaochaihu Decoction+spine pinching)groups, with 5 mice in each group. The mice in other groups except the blank group were modeled for autoimmune MC.After each group was treated for 7 days, the myocardium tissue and serum were collected, and the levels of tumor necrosis factor-alpha(TNF-α),interleukin-1 beta(IL-1β),creatine kinase-MB(CK-MB),cardiac troponin(cTnT),Toll-like receptor 4(TLR4),nuclear factor-kappa B p65(NF-κ-p65),B-cell lymphoma-2(Bcl-2),Bcl-2-associated protein X(Bax),and cysteinyl aspartate-specific protease(caspase-3)were measured. The myocardium sections were observed. The results showed that the Chinese medical therapies outperformed Western medicine in blocking the TLR4/NF-κF p65 pathway and inhibiting apoptosis in the myocardium(P<0.05).The combined treatment group outperformed other groups in lowering the levels of NF-κB p65,TNF-α,Bax,and caspase-3 and elevating the level of Bcl-2(P<0.05).Compared with the Western medicine group,Chinese medical therapies repaired the myocardial cell disarrangement and reduced the infiltration of inflammatory cells in the myocardium, and the combined treatment showed the best performance. In conclusion, modified Xiaochaihu Decoction combined with spine pinching can prevent myocardial inflammation and reduce apoptosis by inhibiting the TLR4/NF-κB signaling pathway. The combination can play a synergistic role in the treatment of MC.
