Effect of Sisheng Jiangzhi Shugan Tang in Lowering Lipid Deposition in Rats with Non-Alcoholic Fatty Liver Disease by Inducing LXRI-ABCA1 Signaling Pathway
Objective To explore the mechanism of Sisheng Jiangzhi Shugan Tang in improving lipid deposition in the liver through rat models with non-alcoholic fatty liver disease(NAFLD)and provide experimental evidence for the treatment of NAFLD with Sisheng Jiangzhi Shugan Tang. Methods A high-fat diet was used to construct rat models with NAFLD,which were divided into a normal group,a model group,a high-dose group of traditional Chinese medicine(TCM),and a low-dose group of TCM.Hemotoxylin-eosin(HE)staining was used to observe the accumulation of oil in the liver, and serum content of TG,TC,LDL-C,HDL-C,TNF-α,and IL-13 was measured.Western blotting was used to detect the effect of Sisheng Jiangzhi Shugan Tang on LXRαTand ABCA1 protein. Results The results of gross and HE staining observation of the liver showed that Sisheng Jiangzhi Shugan Tang significantly reduced lipid accumulation in rat liver, inhibited TNF-αnsecretion(P<0.05),promoted IL-13 secretion(P<0.01),and lowered LDL-C content in rat serum(P<0.05).Sisheng Jiangzhi Shugan Tang increased the expression level of LXRαXand ABCA1 protein(P<0.01).Conclusion Sisheng Jiangzhi Shugan Tang can significantly improve lipid accumulation in rat liver, and the activation of the LXRαXABCA1 signaling pathway to promote lipid efflux may be an important mechanism by which Sisheng Jiangzhi Shugan Tang alleviates NAFLD.
