养血祛风止痛颗粒对头痛与疼痛大鼠模型镇痛及急性血瘀大鼠模型血液流变学参数的影响

摘要

目的 评估养血祛风止痛颗粒对头痛与疼痛模型大鼠的镇痛效果及对血瘀模型大鼠血液流变学参数的影响.方法 急性血瘀大鼠模型试验、大鼠头痛模型试验、大鼠疼痛模型试验各用SPF级SD大鼠 60 只,计算机随机数表法分为空白对照组,模型组,阳性药物对照组,养血祛风止痛颗粒大、中、小剂量组,每组 10 只.各试验组模型完成后按方案给药,血瘀试验采血并测定各组大鼠全血及血浆黏度;头痛试验观察各组尖叫、甩头和后肢抓挠面部的潜伏期、持续期,并记录大鼠 60 min内发生尖叫、甩头和后肢抓挠面部的总次数;疼痛试验记录大鼠疼痛反应时间.结果 急性血瘀模型试验中,与模型组相比,养血祛风止痛颗粒中、大剂量组与复方丹参片阳性药物对照组血浆黏度、全血黏度均显著下降(P<0.05);养血祛风止痛颗粒小剂量组全血黏度无明显变化,差异无统计学意义(P>0.05).头痛与疼痛模型试验中,与模型组相比,养血祛风止痛颗粒中、大剂量组与阳性药物对照组第一时相、第二时相头痛次数均显著降低,潜伏期时长、持续期时长均显著延长;模型组第一时相、第二时相头痛次数显著升高,潜伏期时长、持续期时长显著缩短,差异具备统计学意义(P<0.05).养血祛风止痛颗粒小剂量组与模型对照组第一时相、第二时相头痛次数、潜伏期时长、持续期时长差异无统计学意义(P>0.05).结论 养血祛风止痛颗粒能显著降低头痛与疼痛模型大鼠疼痛应激并改善头痛症状,可降低急性血瘀型模型大鼠全血黏度、血浆黏度,提示养血祛风止痛颗粒具有活血止痛的现代药理作用.

Abstract

Objective To evaluate the analgesic effect of Yangxue Qufeng Zhitong Granules on headache and pain model rats and its impact on hemorheological parameters in blood stasis model rats.Methods Sixty SPF-grade SD rats were used in each of the acute blood stasis model experiment,headache model experiment,and pain model experiment.The rats were randomly divided into a blank control group,a model group,a positive drug control group,and high,medium,and low dose groups of Yangxue Qufeng Zhitong Granules using a random number table method,with 10 rats in each group.After modeling in each experiment,blood was collected for blood stasis testing,and the viscosity of whole blood and plasma was measured in rats from each group.In the headache test,the latency and duration of screaming,head shaking,and hind limb scratching face in each group were observed,and the total number of occurrences of these behaviors within 60 min was recorded.The pain response time of the rats was recorded in the pain test.Results In the acute blood stasis model,compared with the model group,the medium and high dose groups of Yangxue Qufeng Zhitong Granules,as well as the positive drug control group(Compound Danshen Tablets),showed a significant decrease in plasma viscosity and whole blood viscosity(P<0.05).The low dose group of Yangxue Qufeng Zhitong Granules showed no significant change in whole blood viscosity,with the difference not being statistically significant(P>0.05).In the headache and pain model experiments,compared with the model group,the medium and high dose groups of Yangxue Qufeng Zhitong Granules,as well as the positive drug control group,significantly reduced the number of headaches in the first and second phases,while significantly prolonging the latency and duration of symptoms.The model group exhibited a significant increase in the number of headaches in the first and second phases and a significant reduction in latency and duration(P<0.05).There was no statistically significant difference(P>0.05)between the low dose group of Yangxue Qufeng Zhitong Granules and the model control group in terms of the number of headaches in the first and second phases,latency duration,and symptom duration.Conclusion Yangxue Qufeng Zhitong Granules can significantly reduce pain stress and improve headache symptoms in headache and pain model rats.Additionally,it can reduce whole blood viscosity and plasma viscosity in acute blood stasis model rats,indicating that Yangxue Qufeng Zhitong Granules have modern pharmacological effects of promoting blood circulation and relieving pain.
