不同前处理方法对美洲大蠊油脂GC-MS分析影响

目的 分析不同处理美洲大蠊油脂提取物的化学成分,为其开发利用提供实验依据.方法 用超临界CO2萃取法提取美洲大蠊成虫油脂样品经不同处理后制得各供试品,通过色谱-质谱-计算机联用仪进行分离和定性分析,应用色谱峰面积归一化法测定各供试品中各化学成分的相对百分含量并确定美洲大蠊油脂GC-MS分析较优处理方法.结果 美洲大蠊非甲酯处理分离和鉴定出13种成分;酸甲酯化处理分离和鉴定出27种成分;碱甲酯化处理分离和鉴定出42种成分.此三种处理方法分离和鉴定出来的化合物均以烷烃类和脂肪酸酯类化合物为主,但在种类与相对含量上有较大差别.结论 美洲大蠊油脂化合物含多种化合物,其中以烷烃类和脂肪酸及其酯类化合物为主.美洲大蠊油脂GC-MS分析先进行甲酯化处理能明显优化分析结果.
Effects of Different Pretreatment Methods on Periplaneta americana Oil Compounds by GC-MS
Objective To analyze chemical constituents of Periplaneta americana oil extract after different treatments for its development and provide the experimental basis for the uses.Methods Using the supercritical CO2 to extract Periplaneta americana imago oil after different treatments and the samples were separated and qualitative analyzed by GC-MS-DS.Relative percentage content of the chemical constituents in the test samples was determinated by chromatographic peak area normalization method and the better treatment method of Periplaneta americana grease GC-MS analysis was determinated.Results Non-methyl esterification treatment separated and identified 13 components ofPeriplaneta americana; acid methyl esterification treatment separated and identified 26 components; alkali methyl esterification treatment separated and identified 48 components.Alkanes and fatty acid esters were the main compounds by three treatment methods,but there is great difference in the type and relative content.Conclusion Periplaneta americana oil contains a variety of compounds mainly with alkanes and fatty acids esters.It could significantly optimize the analysis results by esterification treatment before GC-MS analysis.
