摘要
目的 为了探究不同培养条件对桑黄菌丝体生长的影响,在短期内培养出质优、含多糖量高的菌丝体.方法 通过观察、对比接种于不同的氮源、碳源培养基中桑黄菌丝体的生长曲线、生长形态、生物量、生理生化等指标,筛选出最适的桑黄菌丝体生长培养基.结果 桑黄生长的最佳液体、固体培养基均为黄豆+玉米的综合培养基:黄豆50 g/L,玉米粒50 g/L,葡萄糖30 g/L,KH2PO43 g/L,MgSO4 0.75g/L,维生素B11 mg/L.结论 最佳单因子结合对桑黄的生物量、多糖积累有协同效应,组合后的发酵体系中生物量达到32 g/L,多糖量达到5.661mg/ml.
Abstract
Objective To explore the influence of different culture conditions on the growth of Phellinus igniarius and to cultivate high-quality mycelia with high content of polysaccharose in the short run.Method Through the observation and comparison over the growth curve and morphology,biomass,and physiological and biochemical indicators of mycelia of Phellinus igniarius in different nitrogen source and carbon source media,the optimum culture medium for the growth of mycelia of Phellinus igniarius was singled out.Results The optimum liquid medium and solid medium for the growth of Phellinus igniarius are both the synthetic medium of corn and soybean,including soybean 50 g/L,niblet 50 g/L,glucose 30 g/L,KH2PO4 3 g/L,MgSO4 0.75 g/L,and Vitamin B1 1mg/L.Conclusion The optimum single factor integration has synergistic effect on the accumulation ofbiomass and polysaccharose,and in the combined fermentation system,the biomass reached up to 32 g/L and polysaccharose reached up to 5.661 mg/ml.
