Exploring the Spleen Strengthening Mechanisms of Atractylodes chinensis and Atractylodes coreana Based on Mitochondrial Autophagy
Objective To study the effects of Atractylodes chinensis and Atractylodes coreana on mitochondrial autophagy expression mediated by the PINK 1-Parkin pathway in the gastric antrum of rats with spleen deficiency,and to explore their spleen strengthening mechanisms and similarities and differences.Methods The experiment was divided into blank group,model group,Atractylodes chinensis group and Atractylodes coreana group.The spleen deficiency model was established using methods such as improper diet,excessive fatigue,and bitter cold diarrhea.Colorimetric method was used to detect the levels of ATP and H2O2 in rat gastric antrum tissue;JC-1 fluorescence probe method was used to detect MMP levels;Western blotting and qRT PCR methods were used to detect the expression of PINK1 Parkin pathway proteins and mRNA in the gastric antrum of rats in each group.Results Compared with the blank group,the model group showed a significant decrease in MMP levels in the gastric antrum(P<0.01),a decrease in ATP levels(P>0.05),and a significant increase in H2O2 levels(P<0.01);The expression levels of PINK1,Parkin,p62 proteins and mRNA were significantly increased(P<0.01 orP<0.05),LC3 mRNA expression was significantly increased(P<0.01),LC3 Ⅱ protein expression was significantly reduced(P<0.01),and the conversion of LC3 Ⅰ to LC3 Ⅱ was reduced(P>0.05).Compared with the model group,the above indicators of the spleen deficiency rats in the Atractylodes chinensis and Atractylodes coreana group showed a downward trend.Conclusion Both Atractylodes chinensis and Atractylodes coreana can improve mitochondrial autophagy function in spleen deficiency rats by regulating the PINK 1-Parkin pathway,exerting a spleen strengthening effect.Among them,Atractylodes coreana has a better regulatory effect than Atractylodes chinensis.
