Study on the Mechanism of Anti-atherosclerosis Effect of Sappanwood by Regulating Immune Inflammatory Reaction Based on Network Pharmacology and its Experimental Verification in vitro
Objective To explore the mechanism of Sappanwood in the treatment of atherosclerosis(AS).Methods The active ingredients of Sappanwood were screened through Herb database,and their targets were predicted by Swiss Target Prediction.The targets of AS were obtained through Gene Cards,DisGeNET and other databases.Cytoscape software was used to construct the"Sappanwood-active ingredient-intersection target"network diagram,the protein interaction network(PPI)of intersection targets was made in the STRING online platform,and the David database was used to perform GO and KEGG enrichment analysis of common targets.The important signaling pathway was verified by cell experiment.Results 443 potential targets of Sappanwood were obtained,of which 78 intersection targets with AS.KEGG analysis showed that key pathways included:HIF-1,AGE-RAGE,Notch signaling pathway,etc.In vitro experiment had shown that Sappanwood ethyl acetate extract can play an anti-atherosclerotic effect by downregulating the expression of Notch1/NICD/Hes1 signaling pathway,upregulating the function of Tregs,inhibiting the migration of smooth muscle cells,promoting the expression of anti-inflammatory factors such as IL-10 and TGF-β1,and reducing the expression of inflammatory factors such as IL-6 and IL-12.Conclusion Sappanwood plays an anti-AS role by regulating Notch signaling pathway and other pathway.
