摘要

目的 建立超高效液相色谱-四极杆-静电场轨道阱高分辨质谱(UPLC-QE-Orbitrap-MS),系统开展中药复方四物汤化学成分的表征与鉴定.方法 采用BEH C18色谱柱(2.1 mm × 100 mm,1.7 µm),流动相为0.1%甲酸水-乙腈,流速为0.5 mL/min,柱温为30 ℃,在正、负离子模式下采用Auto MS/MS进行数据采集.结果 通过二级质谱裂解碎片解析以及文献分析等,从复方四物汤中共鉴定或初步推导了 47种化合物,包括5种有机酸、3种酚类、12种黄酮类、6种苯丙素类、15种萜类、2种倍半萜和4种生物碱.结论 建立的UPLC-QE-Orbitrap-MS能快速、准确、较全面地鉴定复方四物汤的化学成分,为其药效物质基础研究提供参考依据.

Abstract

Objective To establish high resolution mass spectrometry with ultra-high performance liquid chromatography(UPLC-QE-Orbitrap-MS)and systematically represent and identify the chemical composition of TCM compound Siwu Tang.Methods Data acquisition by BEH C18 column(2.1 mm× 100 mm,1.7 μm),0.1%formic acid water-acetonitrile,flow rate was 0.5 mL/min,column temperature was 30℃,and Auto MS/MS in positive and negative ion mode.Results 47 compounds,including 5 organic acids,3 phenols,12 flavonoids,6 phenylpropanoids,15 terpenes,2 sesquiterpenes and 4 alkaloids were identified or preliminarily derived from the compound Siwu Tang through secondary mass spectrometry fragmentation analysis and literature analysis.Conclusion The established UPLC-QE-Orbitrap-MS can quickly,accurately and comprehensively identify the chemical components of compound Siwu Tang,providing a reference basis for the study of its pharmacological substance basis.
