Cytometabonomic Study of Anti-inflammatory Activity of Dehydrodiisoeugenol
Objective To explore the anti-inflammatory activity and mechanism of dehydrodiisoeugenol. Methods By exploring the changes of inflammatory factors in RAW264.7 cells induced by LPS,the drug had anti-inflammatory effect to verify whether it had anti-inflammatory effects, and UPLC-Q-Exactive Orbitrap-MS was used to analyze metabolomics to obtain differential metabolites between groups and analyze related metabolic pathways. Results Dehydrodiisoeugenol significantly inhibited the content of three inflammatory factors:IL-6,TNF-α and IL-1β;After metabolomics screening,43 endogenous differential metabolites were obtained, mainly phosphatidylcholine and free fatty acids. After enrichment analysis, three main metabolic pathways were obtained:arginine biosynthesis, alanine, aspartic acid and glutamic acid metabolism, and glycerol phospholipid metabolism. Conclusion Dehydrodiisoeugenol mainly has obvious inhibitory effects on inflammatory factors, and the play of its inhibitory effect may be related to amino acid metabolism and lipid metabolism.
