去氢二异丁香酚抗炎活性的细胞代谢组学研究

目的 研究去氢二异丁香酚的抗炎活性及作用机制。方法 研究药物对经LPS诱导后的RAW264。7细胞中炎症因子变化情况,验证其是否具有抗炎作用;采用超高效液相色谱-四极杆静电场轨道阱高分辨质谱技术进行代谢组学分析,获取组间差异代谢产物,分析相关的代谢通路。结果 去氢二异丁香酚明显抑制了 IL-6、TNF-α、IL-1 β三种炎症因子的含量;经代谢组学筛选得到内源性差异代谢物43个,主要为磷脂酰胆碱和游离脂肪酸等成分。通过富集分析筛选后得到精氨酸生物合成,丙氨酸、天冬氨酸和谷氨酸代谢以及甘油磷脂代谢三条主要代谢通路。结论 去氢二异丁香酚对炎症因子具有明显的抑制作用,其抑制作用的发挥可能与氨基酸代谢和脂质代谢有关。
Cytometabonomic Study of Anti-inflammatory Activity of Dehydrodiisoeugenol
Objective To explore the anti-inflammatory activity and mechanism of dehydrodiisoeugenol.Methods By exploring the changes of inflammatory factors in RAW264.7 cells induced by LPS,the drug had anti-inflammatory effect to verify whether it had anti-inflammatory effects,and UPLC-Q-Exactive Orbitrap-MS was used to analyze metabolomics to obtain differential metabolites between groups and analyze related metabolic pathways.Results Dehydrodiisoeugenol significantly inhibited the content of three inflammatory factors:IL-6,TNF-α and IL-1β;After metabolomics screening,43 endogenous differential metabolites were obtained,mainly phosphatidylcholine and free fatty acids.After enrichment analysis,three main metabolic pathways were obtained:arginine biosynthesis,alanine,aspartic acid and glutamic acid metabolism,and glycerol phospholipid metabolism.Conclusion Dehydrodiisoeugenol mainly has obvious inhibitory effects on inflammatory factors,and the play of its inhibitory effect may be related to amino acid metabolism and lipid metabolism.
