Ameliorating Effect of Qianyang Yuyin Decoction and Valsartan on Gut Microbiota Dysbiosis in Spontaneously Hypertensive Rats
Objective To explore the regulatory effect of Qianyang Yuyin decoction combined with valsartan on the gut microbiota dysbiosis in spontaneously hypertensive rats(SHRs).Methods 32 male SHR rats of 12 weeks old were randomly divided into modelgroup(MG),valsartan group(XG),Qianyang Yuyingroup(ZG)and Qianyang Yuyin combined valsartan group(ZXG),as well as 8 male Wistar-Kyoto rats(WKYs)of the same age were as control group(CG).The systolic blood pressure(SBP)and the diastolic blood pressure(DBP)of rats were monitored by noninvasive blood pressure measurement of tail artery before and after 8 weeks of treatment,respectively.After 8 weeks of administration,the fecal DNA was extracted and 16S rRNA gene was sequenced and the results were analyzed.The systolic blood pressure(SBP)and the diastolic blood pressure(DBP)of rats were monitored by noninvasive blood pressure measurement of tail artery before and after 8 weeks of treatment,respectively.Results The anti-hypertensive potency of herb-drug combination intervention was much better than Qianyang Yuyin decoction.The relative abundances of Coprobacillus,Lactococcus,Phascolarctobacterium,Eubacterium,Elusimicrobium and Enterococcus were reduced in MG group,however,the relative abundances of Coprobacillus and Eubacterium were reversed after Qianyang Yuyin decoction treatment;and those of Phascolarctobacterium,Enterococcus,Eubacterium,Lactococcus were reversed after herb-drug combination treatment.PICRUSt function prediction showed that Qianyang Yuyin decoction combined with valsartan could restore the disordered gut microbiota composition and metabolism.Conclusion Qianyang Yuyin decoction combined with valsartan can exert anti-hypertensive effect by restoring gut microbiota dysbiosis in SHRs,which provide theoretical basis for herb-drug combination.
