Protective Effect and Mechanism of Cirsium tianmushanicum Shih Extract on Chronic Alcoholic Liver Injury in Mice
Objective To study protective effect and mechanism of Cirsium tianmushanicum Shih extract(CSE)on chronic alcoholic liver injury in mice.Methods 36 healthy C57/BL6 mice were randomly divided into normal,model,BIF,CSE-1,CSE-m and CSE-h groups,6 mice each group.Physiological saline was given to normal group while 50%ethanol was given to the other groups.1h later,mice in normal group and model group were given physiological saline,and mice in the other groups were given corresponding drugs.30 d later,the liver indexand the serum and liver biochemical indexes of mice were detected.Pathological changes of liver and small intestine,lipid accumulation in the liver and apoptosis of liver cells were respectively detected.The expression levels of proteins on the Bax/Bcl-2/Caspase-3 signal path were detected by Western Blotting.Results Compared with model group,the liver index,the activitiy of AST,ALT and the content of MDA and TG of Cirsium tianmushanicum Shih extract group mice were significantly decreased(P<0.01)while the activities of GSH were significantly increased(P<0.01).The morphology of liver and small intestine was significantly improved in the treatment group.It was showed that the apoptosis in liver tissue of each treatment group was significantly decreased.The expression of Bax,Cyt-C,Cleaved Caspase-3 proteins decreased(P<0.01),while the expression of Bcl-2 protein increased(P<0.01),and the expression of Caspase-3 protein had no difference.(P>0.01).Conclusion Cirsium tianmushanicum Shih extract can protect the chronic alcoholic liver injury,which may be due to anti-oxidation,regulation of lipid metabolism and inhibition of Csapase-3/Bax/Bcl-2 apoptosis pathway.
