Study on Clarifying Technology of Linggui Zhugan Oral Liquid and its Anti-fatty Liver Effect
Objective To optimize the clarifying process of Linggui Zhugan oral liquid and to study the anti-fatty liver effect.Methods The effects of water extraction and alcohol precipitation,clarifier,high speed centrifugation,membrane filtration and heat treatment on the content and clarity of glycyrrhizic acid in Linggui Zhugan oral liquid were compared.The body weight of mice was measured,and the liver lipid droplets were studied by hematoxylin-eosin staining and oil red O staining.Results After comparing different clarifying processes of Linggui Zhugan oral liquid,it was concluded that ZTC1+1 clarifying agent method had the highest comprehensive score,so ZTC1+1 clarifying agent method was chosen as the best clarifying process of Linggui Zhugan oral liquid.The weight of mice in the model group was higher than that in the blank group,and the weight of mice decreased significantly after the intervention of oral liquid,especially in the high-dose group.The results of hematoxylin-eosin staining showed that compared with the model group,the lipid vacuole and hepatocyte swelling in the treatment group were significantly reduced.The results of oil red O staining showed that lipid deposition in the liver tissue of mice in the treatment group was significantly reduced compared with that in the model group.Conclusion The best clarifying process of Linggui Zhugan oral liquid is ZTC1+1 clarifying agent method,the dosage of clarifying agent is 4％:2％,the concentration of liquid is 1∶4,and the flocculation temperature is 70℃.Linggui Zhugan oral liquid can reduce body weight and liver fat droplet deposition in mice,and preliminatively explore its anti-fatty liver effect.
