基于AHP-熵权法优选降糖灵口服液提取纯化工艺

目的 优选降糖灵口服液的提取纯化工艺,为其工业生产提供参考.方法 基于AHP-熵权法给总多糖量、固体总量、葛根素、毛蕊异黄酮葡萄糖苷四个评价指标进行主客观赋权.采用L9(34)正交设计对提取的料液比、提取时间、提取次数进行考察,以四个评价指标的综合评分优选最佳提取工艺.采用Box-Behnken响应面设计对纯化工艺的浸膏相对密度、醇浓度、静置时间进行考察,以四个评价指标的综合评分优选最佳纯化工艺.结果 降糖灵口服液的最佳提取工艺为加8倍量水,提取3次,每次1h;最佳纯化工艺为将浸膏浓缩至相对密度1.15,加95%乙醇至含醇量达65%,静置12h.3批验证试验结果无明显差异.结论 优选的降糖灵口服液提取纯化工艺稳定可靠,可以为大生产提供理论依据.
Optimization of Extraction and Purification for Jiangtangling Oral Liquid on AHP-entropy Weight Method
Objective To optimize the extraction and purification process of Jiangtangling oral liquid to provide reference for industrial production.Methods Based on AHP-entropy weight method,the four evaluation indexes of total polysaccharide content,total solid content,puerarin and calycosin-7-glucoside were empowered subjectively and objectively.Orthogonal design was used to investigate the material-to-liquid ratio,extraction time and extraction duration in the water extraction process,and the best extraction process was optimized by the comprehensive score of four evaluation indexes.Box-Behnken design was used to investigate the effect of the relative density of the extract,volume fraction of ethanol for precipitation,and precipitation duration in the ethanol precipitation process,and the best purification process was optimized by the comprehensive score of four evaluation indexes.Results The best extraction process of Jiangtangling oral liquid was material-to-liquid ratio of 1∶8,extraction for 1 h with a total of 3 times;the best purification process was to concentrate the extract to relative density of 1.15,add 95%ethanol to 65%alcohol and stand for 12 h.There was no significant difference in the 3 batches of validation tests.Conclusion The preferred extraction and purification process of Jiangtangling oral liquid is stable and reliable,which can provide a theoretical basis for industrial production.
