Study on the Mechanism of Asiaticoside Improving Colitis in TNBS Model Mice
Objective To investigate the effect and potential mechanism of asiaticoside(AS)on CD-like enteritis.Methods Twenty-four male C57BL/6J mice aged 6-8 weeks were selected and randomly divided into normal group(WT group),control group(TNBS group),experimental group(TNBS+AS group)and positive control group(WT+AS group).DAI score,weight change,colon length,inflammation score,colon macro score and expression level of inflammatory factors were used to evaluate the effects of AS on intestinal inflammation in TNBS mice.The effects of AS on intestinal barrier function in TNBS mice were evaluated by FITC-Dextran,16S rDNA level,Bacterial translocation and TEER.The effects of AS on the proliferation and differentiation of intestinal stem cells in TNBS mice were detected by immunohistochemistry and immunofluorescence assay.The effects of AS on Neurog3 and SPDEF,which regulate intestinal stem cell differentiation,were detected by Western Blotting and RT-qPCR.Results AS could significantly relieve intestinal inflammation in TNBS mice(P<0.05).At the same time,intestinal barrier dysfunction of TNBS mice was significantly improved after AS intervention(P<0.05).Immunohistochemical and fluorescence experiments showed that AS significantly promoted the proliferation and differentiation of intestinal stem cells in TNBS mice(P<0.05).Western Blotting assay and RT-qPCR results showed that AS increased the expression levels of Neurog3 and SPDEF(P<0.05).Conclusion AS can effectively improve colitis in TNBS mice by promoting the proliferation and differentiation of intestinal stem cells.
