基于UPLC-Q-TOF-MS/MS的黄精奶制前后化学成分变化分析

目的 采用UPLC-Q-TOF-MS/MS建立黄精及奶黄精成分分析方法,对黄精炮制前后化学成分进行分析鉴定。方法 以超声提取法对黄精及奶黄精进行样品前处理,利用超高效液相色谱法(UPLC)结合四级杆飞行时间质谱(Q-TOF-MS/MS),采用电喷雾离子源,以正、负离子模式进行扫描,结合SCIEX OS V 2。1中药化合物数据库、PubMed、中国知网等数据库对黄精奶制前后的化学成分进行表征。结果 从黄精及奶黄精中鉴定化学成分92个,其中,黄精中化学成分79个、奶黄精化学成分59个,共有成分46个,奶制后黄精黄酮及氨基酸类成分显著减少,而甾体皂苷类成分发生明显的异构。结论 利用UPLC-Q-TOF-MS/MS技术有效分析了黄精及其奶制品化学成分,黄精及奶制品之间化学成分差异显著,为黄精的质量控制和资源开发提供参考。
Analysis on Chemical Components Changes of Polygonati Rhizoma Before and After Processing with Milk Based on UPLC-Q-TOF-MS/MS
Objective To establish a method for the composition analysis of Mongolian medicine Polygonati Rhizoma(P.Rhizoma)and milk processing P.Rhizoma by UPLC-Q-TOF-MS/MS.Methods The samples were processed by ultrasonic extraction method.ACQUITY UPLC©HSS T3 was used as stationary phase,0.1%formic acid-acetonitrile and 0.1%formic acid-water were used as mobile phase elution system,electrospray ionization source was used to scan in positive and negative ion mode,and SCIEX OS workstation was used to extract and process the chromatography.The chemical constituents of the two samples were characterized based on SCIEX OS V 2.1 Chinese Medicine Compound database,PubMed,CNKI and other databases.Results A total of 92 chemical components were identified by IDA scanning,including 79 chemical components in P.Rhizoma and 59 chemical components in milk processing P.Rhizoma,46 components were common component.After processing,there was a significant reduction in the composition of flavonoids and amino acids,while the component of steroidal saponins showed obvious heterogeneity.Conclusion UPLC-Q-TOF-MS/MS technology can be used to effectively analyze the chemical composition of Mongolian medicine P.Rhizoma and its processed products,chemical compositions exhibit significant disparities between them,which provides a reference for the quality control and resource development of P.Rhizoma.
