THE FERROPTOSIS MECHANISM ON COLON CANCER HCT116 CELLS INDUCED BY COMPOUND GEGEN DECOCTION
To investigate the effect of compound Gegen decoction on the growth of colon cancer HCT116 cells and its mechanism,the effect of compound Gegen decoction on the growth of HCT116 cells was investigated by MTT,clone formation assay and morphological analysis,and reactive oxygen species(ROS)were analyzed by kit and fluorescence microscopy,and then its mechanism was verified and explored by western blotting assay.The results showed that compared with the vehicle group,compound Gegen decoction could significantly inhibit the growth of HCT116 cells in a concentration and time dependent manner;Mechanically,compound Gegen decoction could significantly increase reactive oxygen,and up-regulate the expression of ferroptosis-related protein PTGS2 and ACSL4,and down-regulate the expression of ferroptosis-related protein GPX4.These results suggest that compound Gegen decoction may inhibit the growth of HCT116 cells by inducing ferroptosis.
