An Effective Method for Measuring the Content of Free Sodium Hyaluronate
A method for the determination of free sodium hyaluronate by ultraviolet spectrophotometry was established.The application of free sodium hyaluronate is limited due to its solubility in water.The use of crosslinking agents to synthesize crosslinked sodium hyaluronate overcomes the defects of natural sodium hyaluronate,but it is difficult to disperse in aqueous solution,and other water-soluble substances in the crosslinked network need to be dissolved before they can be released,thus reducing the effectiveness of measuring the content of free sodium hyaluronate.On the basis of the existing research methods,the measurement results are greatly improved by changing the dissolution mode and filtration mode.In the range of 0~50μg/mL,the linear relationship between sodium hyaluronate content and absorbance was good,and the correlation coefficient was 0.99982.The minimum detection limit was 0.008μg/mL.The limit of quantification was 0.025μg/mL.For precision test,RSD was 0.02%(n=10);Repeatability test,RSD was 4.3%(n=10);The average recovery rate was 94%(n=10).The method has the advantages of high accuracy,good stability,high sensitivity and good repeatability.And it has more specific and applicable.It can be used to measure the content of free sodium hyaluronate in various types of crosslinked sodium hyaluronate gel products.
