HPLC法测定泡吴茱萸中柠檬苦素、吴茱萸碱和吴茱萸次碱含量

目的:建立用高效液相色谱法(HPLC)测定泡吴茱萸中柠檬苦素、吴茱萸碱、吴茱萸次碱三种成分的含量.方法:色谱柱为Eclipse XDB C18(4.6 × 250mm,5μm),流动相:A乙腈,B甲醇,C 0.05%磷酸溶液,各流动相配比为30∶20∶50,流速:1.000mL·min-1,检测波长:215nm,柱温:30℃.结果:柠檬苦素、吴茱萸碱、吴茱萸次碱三者依次在 0.1303~1.6291 μg(r=0.9995)、0.0500~0.6251 μg(r=1.0000)、0.0354~0.4429μg(r=0.9999)范围内与其峰面积有良好的线性关系,且三者的平均加样回收率和RSD值分别为 100.11%(RSD=0.7%)、100.74%(RSD=1.4%)和 99.39%(RSD=1.3%).结论:该方法操作简单,结果稳定,有着高准确度,适用于泡吴茱萸中柠檬苦素、吴茱萸碱、吴茱萸次碱含量的测定.
HPLC Determination of Limonin,Codiamine and Rutacarpine in Processed Evodia Rutaecarpa(Juss.)Benth
Objective:To establish a HPLC method for the determination of limonin;codiamine and rutacarpine in processed Evodia rutaecarpa(Juss.)Benth.Methods:The analysis was performed on Eclipse XDB C18(4.6 x 250mm,5 μm)column.The mobile phase was acetonitrile-methanol-0.05%phosphoric acid solution(30∶20∶50)at the flow rate of 1.000 mUmin.The detection wavelength was 215 nm and column temperature was set at 30 ℃.Results:The linear ranges of limonin;codiamine and rutacarpine were 0.1303~1.6291 μg(r=0.9995),0.0500~0.6251 μg(r=1.0000),0.0354~0.4429 μg(r=0.9999),respectively.The average recovery rates were 100.11%(RSD=0.7%),100.74%(RSD=1.4%),99.39%(RSD=1.3%).Conclusion:This method is simple,accurate and proper for simultaneous determination of limonin,codiamine and rutacarpine in processed Evodia rutaecarpa(Juss.)Benth.
