Optimization of Enzymatic Extraction of Collagen from Eggshell Membrane
Collagen has the functions of promoting wound healing and hemostasis.At present,the sources of collagen on the market are done-hide gelatin and various animal skin adhesives.Collagen can also be extracted from eggshell membrane,but there are few related studies.The waste eggshell was used as raw material,firstly soaked in hydrochloric acid,the shell membrane was separated,the protein was purified with sodium hydroxide,and the collagen in the waste eggshell membrane was extracted by pepsin enzymolysis and salting out method.The concentration of pepsin,pH,time and temperature were investigated for single factor test,and then the optimal extraction process was obtained by L9(34)orthogonal test.The content of collagen in eggshell film was determined by UV spectrophotometry.The results showed that the extraction rate of collagen from egg shell membrane was 30.41%under the conditions of 40 ℃,pH 1.7,30 h enzymolysis reaction time and 10%enzyme dosage.Compared with related studies,the extraction rate increased by 0.361%.
