Study on Optimization of Extraction Technology and Antioxidant Activity of Polysaccharide from Aromatic Tobacco
Objectives:The purpose of this study was to optimize the extraction process of tobacco polysaccharides and to further explore their antioxidant activities.Methods:1.The effects of ultrasonic power,ultrasonic time and extraction temperature on the extraction rate of tobacco polysaccharides were studied.And the optimal extraction conditions were obtained by response surface optimization.DPPH radical scavenging,ABTS radical scavenging and o-diazepine scavenging were used to analyze the scavenging ability of free radicals in vitro.Results:The optimal ultrasonic extraction conditions were determined as follows:time 80min,temperature 70℃,power 70W.The extraction rate was 3.02%.When the concentration of aromatic tobacco polysaccharide was the range of 0~5 mg/mL,the scavenging rate of hydroxyl free radical increased continuously,reaching 71.26%,and the IC50 value was 2.319 mg/mL.The removal rate of DPPH increased and then decreased with IC50 of 4.247 mg/mL when the concentration of tobacco polysaccharide increased in the range of 0~10 mg/mL,the ABTS clearance rate increased with the increase of polysaccharide solution concentration.The IC50 value was 6.418 mg/mL.
