超高效液相色谱法同时测定糕点中的安赛蜜、苯甲酸、山梨酸、糖精钠、脱氢乙酸的方法研究

建立超高效液相色谱法同时测定糕点中安赛蜜、苯甲酸、山梨酸、糖精钠和脱氢乙酸 5 种食品添加剂含量的方法.样品经硫酸锌与氢氧化钠溶液(加氢氧化钠溶液至pH值为 7.5)沉淀蛋白质,超声提取,使用Waters ACQUITY UPLC BEH C18 色谱柱,以甲醇+0.02 mol/L乙酸铵溶液(用氨水调节Ph值至7.2~7.6)=3+97 为流动相进行等度洗脱,流速 0.2 Ml/min,柱温 35℃,采用二极管阵列检测器,检测波长230nm,用外标法定量.5 种食品添加剂在 1~50 μg/Ml范围内线性关系良好,相关系数r≥0.9999,检出限为0.591 mg/kg~0.906 mg/kg,平均加标回收率为 95.6%~100.1%,相对标准偏差为 0.12%~1.40%,在 7.6 min之内5 种食品添加剂能够完全分离.该方法简单、快速、回收率高、精密度好,适用于糕点中 5 种食品添加剂的同时检测.
Research on Simultaneous Determination of Acesulfame,Benzoic Acid,Sorbic Acid,Saccharin Sodium,and Dehydroacetic Acid in Pastries Using Ultra-high Performance Liquid Chromatography
Establish a method for simultaneous determination of five food additives,namely acesulfame,benzoic acid,sorbic acid,saccharin sodium,and dehydroacetic acid,in pastries using ultra-high performance liquid chromatography.The sample was precipitated with zinc sulfate and sodium hydroxide(adding sodium hydroxide solution to pH 7.5)to precipitate protein,then use ultrasound for extraction.The sample was separated by Waters ACQUITY UPLC BEH C18 column,and it was eluted equally using methanol+0.02mol/L ammonium acetate solution(adjusted to pH 7.2~7.6 with ammonia water)=3+97 as the mobile phase.Set the flow rate to 0.2mL/min and the column temperature to 35℃.The sample was detected using a diode array detector with a detection wavelength of 230nm,and quantified using an external standard method.Five kinds of food additives had good linear relationships in the range of 1~50 μg/mL,and the correlation coefficients r≥0.9999.The limits of detection were 0.591mg/kg~0.906mg/kg.The average recoveries of five kinds of food additives were 95.6%~100.1%,with the relative standard deviations of 0.12%~1.40%.Five kinds of food additives can be completely separated within 7.6 minutes.This method is simple,fast,with high recovery rate and good precision,and is suitable for the simultaneous detection of five food additives in pastries.
