Quercetin inhibits CD38/ERK pathway-mediated inflammatory response of alveolar epithelial cell
Objective To investigate the potential relationship between the expression of CD38 and inflammatory factors in peripheral blood leukocytes of patients with pulmonary infection,and the inhibitory mechanism of quercetin on LPS-induced in-flammatory response of alveolar epithelial cells.Methods Blood samples of patients with pulmonary infection admitted to Jiangxi Provincial People's Hospital from May 2023 to December 2023 were collected as the infection group(n=50),and blood samples of healthy subjects in our hospital during the same period were selected as the control group(n=50).Flow cytometry was used to de-tect the levels of CD38 and inflammatory factors in peripheral blood cells.Bioinformatics predicted the relationship between quercetin and CD38,CCK-8 method was used to detect the effects of LPS and quercetin on the viability of MLE-12 cells,and western blot was used to detect the expression levels of CD38,IL-18,TNF-α and MAPK pathway-related proteins.Results The expression levels of CD38 on peripheral white blood cells and lymphocytes in pulmonary infection group were significantly higher than those in control group(P<0.05),and the levels of IL-6(P<0.01)and IL-10(P<0.05)in pulmonary infection group were also signif-icantly higher than those in control group.The results of cell experiment showed that CD38,IL-18,TNF-α and P-ERK/ERK levels in LPS+quercetin group were significantly decreased compared with LPS group(P<0.05).Conclusion CD38 expression is up-reg-ulated in peripheral blood leukocytes of pulmonary infection,mainly in lymphocytes.CD38 promotes the release of inflammatory factors in peripheral blood,which is one of the mechanisms of pulmonary inflammation.CD38 is a potential target of quercetin regulated inflammatory responses,and quercetin can reduce the release of LPS-induced inflammatory factors IL-18 and TNF-α in MLE-12 cells by inhibiting the CD38/ERK pathway.
