Objective:To investigate the effect of canopy FGF signal regulator 3(CNPY3)on the proliferation and invasion of colorectal cancer cells and its molecular mechanism.Methods:The expression of CNPY3 in colorectal cancer tissues was analyzed by the TCGA database and Western blot method.The effect of CNPY3 on the proliferation was detected by CCK8 and colony formation.The effect of CNPY3 on the invasion of colorectal cancer cells was detected by Transwell assay.The expression of E-cadherin,N-cadherin,and vimentin proteins related to the epithelial-mesenchymal transition(EMT)signaling pathway in colorectal cancer cells was determined by Western blot.Results:TCGA database analysis showed that the expression level of CNPY3 in colorectal cancer tissues was significantly higher than that in the adjacent normal colorectal tissues(P<0.01).The results of Western blot showed that the expression level of CNPY3 in the resected colorectal cancer specimens was significantly higher than that in the adjacent normal colorectal tissue specimens(P<0.01).CCK8 assay and colony formation assay showed that CNPY3 knockdown significantly suppressed the proliferation of colorectal cancer cells(P<0.01).Transwell assay showed that CNPY3 knockdown significantly inhibited the invasion ability of colorectal cancer cells(P<0.01).Western blot results showed that after CNPY3 knockdown,the expression of E-cadherin in colorectal cancer cells increased,and the expression of N-cadherin and vimentin decreased(P<0.01).Conclusions:CNPY3 is highly expressed in colorectal cancer tissues and may regulate the proliferation and invasion of colorectal cancer cells through EMT signaling pathway.
