Effects of Yulangsan polysaccharides on BDNF pathway in the dentate gyrus of the hippocampus of mice with chronically stressed

Accumulating evidence indicates that the Brain-derived neurotrophic pathway play important roles in depression.Previous studies in our laboratory have demonstrated that YLSPA produced antidepressant-like action in Kunming mice with various mouse models of behavioral despair (FST and TST). The present study aimed to examine whether YLSPA could affect the chronic unpredictable mild stress (CUMS)-induced depression in mice. The mechanism(s) underlying the antidepressant-like action was investigated by measuring brain-derived neurotrophic factor (BDNF) ,its receptor(TrkB) ,the relevant factor of pathway CREB and pCREB using Immunohistochemistry.CUMS, being lasted for 3 weeks, caused depression-like behavior in mice, as indicated by the significant decrease in sucrose consumption and spontaneous motor activity. Whereas CUMS produced depressive-like behaviors and decreased the expression of BDNF , TrkB (p < 0.05 or p <0.01), accompanied by a decreased expression of both CREB and pCREB in the dentate gyrus region of the hippocampus (p < 0.05 or p <0.01). On day 21, CUMS resulted in a decreased number of apoptosis cells and the level of bax in the dentate gyrus zone (p < 0.05 or p <0.01) while taken an increased in the expression of Bcl-2.Daily intragastric administration of YLSPA(300,600 mg/kg) during three weeks of CUMS significantly suppressed behavioral and biochemical changes induced by CUMS.The results suggest that the antidepressant-like action of YLSPA is likely mediated by modulating the function of increasing the expression of BDNF in brain tissues and related factors.
