摘要
本研究在优化前处理条件和色谱分离的基础上,建立了高效液相色谱法(HPLC)测定工业大麻中大麻二酚(CBD)和四氢大麻酚(THC)的检测方法.样品经优化提取剂、流动相及色谱条件,最终选择用95%乙醇提取,乙腈-水(68:32,v/v)为流动相,采用C18色谱柱进行分离,柱温35 ℃,流速1.0 mL/min,进样体积10µL,检测波长220 nm.结果表明,该方法在0.02~0.10 mg/mL浓度范围内具有良好的线性关系,相关系数(r)均大于0.998.方法检出限(LOD,S/N=3)和定量限(LOQ,S/N=10)分别为:CBD的检出限为0.000 1 g/100g、定量限为0.000 3 g/100g;THC的检出限为0.000 6 g/100 g、定量限为0.001 8 g/100 g.在已知样品中加入0.5 mg/kg、50mg/kg和200 mg/kg三个浓度水平的CBD和THC标准溶液,进行加标回收试验,CBD的回收率为92%~101%,THC的回收率为80%~102%.该方法准确性、灵敏性和稳定性良好,能够满足工业大麻中CBD和THC的检测要求.
Abstract
Based on the optimization of pretreatment conditions and chromatographic separation,a High-performance liquid chromatography HPLC method for the determination of cannabidiol(CBD)and A9-tetrahydro-cannabinol(THC)in industrial hemp was developed.The samples were extracted with 95%ethanol,acetonitrile-water(68:32,v/v)as the mobile phase,and separated by C18 column at 35 ℃ with a flow rate of 1.0 mL/min,the in-jection volume was 10 μL and the detection wavelength was 220 nm.The results show that the method has a good linear relationship in the range of 0.02-0.10 mg/mL,and the correlation coefficients(r)are all greater than 0.998.The detection limits(LOD,S/N=3)and quantitative limits(LOQ,S/N=10)were 0.0001 g/100 g and 0.0003 g/100 g for CBD,0.0006 g/100 g and 0.0018 g/100 g for THC,respectively.The recovery rates of CBD and THC were 92%-101%and 80%-102%,respectively when the negative samples were added with 0.5 mg/kg,50 mg/kg and 200 mg/kg.The method has good accuracy,sensitivity and stability,and can meet the detection requirements of CBD and THC in industrial hemp.
