植物生长延缓剂B9对不同品种马铃薯试管薯形成的影响

为探究植物生长延缓剂B9对马铃薯试管大薯的影响,以不同品种的马铃薯脱毒苗为试验材料,在光照强度2 000 Lx、光周期16 h·d-1,培养温度 22℃的培养条件下,以MS培养基为基础培养基,分别设置4%、6%、8%、10%浓度的蔗糖,以3%浓度蔗糖作对照,对比不同蔗糖浓度诱导效果后,确定诱导蔗糖浓度,设置5个B9浓度2 mg·L-1、4 mg·L-1、6 mg·L-1、8 mg·L-1、10 mg·L-1,测定试管薯的平均薯重、最大薯重,统计大薯率,测定试管薯粗蛋白、还原糖与维生素C.结果表明:适合诱导马铃薯试管大薯的培养基为MS+蔗糖8%+B9(6 mg·L-1),诱导30 d,隆丰黑美人、费物瑞它、尤金、延薯13四个品种的平均单薯重分别为0.78 g、0.76 g、0.64 g、0.47 g,最大薯重分别达到0.91 g、0.84 g、0.78 g、0.68 g.
Effect of plant growth retarder B9 on microtuber formation of different varieties of potato
In order to investigate the effect of plant growth retarder B9 on potato in vitro,different varieties of potato virude-free seedlings were used as experimental materials.Under the culture condi-tions of light intensity 2000 Lx,photcycle 16 h·d-1,culture temperature 22℃,MS medium was used as the base medium,and sucrose concentrations of 4%,6%,8%and 10%were set,respectively.After comparing the induction effect of different sucrose concentrations with 3%sucrose concentration,the induced sucrose concentration was determined.Six B9 concentrations of 2mg·L-1,4mg·L-1,6mg·L-1,8mg·L-1 and 10mg·L-1 were set up to determine the average and maximum potato weight of test-tube potato and calculate the large potato rate.Determination of crude protein,reducing sugar and vita-min C in vitro.The results show that:The medium suitable for inducing large potato in vitro was MS+8%sucrose+B9(6mg·L-1).After 30 days of induction,the average single potato weight of Longfeng Black Beauty,Feuritar,Yougin and Yantua 13 varieties was 0.78g,0.76g,0.64g and 0.47g,respec-tively.The maximum potato weight was 0.91g,0.84g,0.78g and 0.68g,respectively.
