384孔PCR仪温度参数校准方法探讨

PCR技术作为一项革命性的分子生物学工具,在基因分析、病原体检测、药物研发、遗传学研究等领域发挥着重要作用.384 孔PCR仪作为高通量PCR仪的代表,具备一次性处理多个样本的能力.由于现有的校准规范主要针对96 孔,使用384 孔时存在校准困难和准确性较差的问题.因此,本文通过对两者进行对比,提出了 384 孔PCR仪的校准布点方式,并进行试验验证.结果表明:采用该校准布点方式具有有效性.
Research on Calibration Method of Temperature Parameters for 384-hole Polymerase Chain Reaction Analyzer
PCR technology,as a revolutionary molecular biology tool,plays an important role in gene analysis,pathogen detection,drug development,genetic research and other fields.As a representative of high-throughput PCR instrument,the 384 well PCR instrument has the ability to process multiple samples at one time.Because the existing calibration specification is mainly for 96 holes,there are problems of calibration difficulty and poor accura-cy when using 384 holes.Therefore,through the comparison of the two,this paper puts forward the calibration lay-out method of 384-well PCR instrument,and carries out the test verification.The experiment shows that this meth-od is effective.
