3首活血化瘀方对寒凝血瘀模型大鼠红细胞膜蛋白组成研究

目的 采用SDS-PAGE法获得红细胞膜蛋白表达谱,用于寻找3首活血化瘀方对寒凝血瘀模型大鼠红细胞膜蛋白组成的影响.方法 80只Wistar大鼠,分为空白组、寒凝血瘀模型组、补阳还五汤高低剂量组、少腹逐瘀汤高低剂量组、丹参饮高低剂量组,随机分为8组,每组10只.采用冰水浸泡和盐酸肾上腺素注射法复制寒凝血瘀模型,造模15d.各给药组1次/d.末次造模后禁食不禁水12h.采用SDS-PAGE法分析红细胞膜蛋白组成变化.结果 与空白组比较,寒凝血瘀模型组红细胞膜上带1、带2、带3和带7蛋白百分含量显著降低(P<0.05).与模型组比较,少腹逐瘀汤可显著升高红细胞膜上带1蛋白、带2蛋白、带3蛋白、带4.1蛋白和带5蛋白的百分含量(P<0.05);丹参饮可显著升高红细胞膜上带1蛋白和带7蛋白的百分含量(P<0.05);补阳还五汤虽没有显著作用,但是有升高的趋势.结论 少腹逐瘀汤和丹参饮可以不同程度的纠正寒凝血瘀模型大鼠红细胞膜的膜蛋白组成.
Study of erythrocyte membrane proteins of the cold stasis rats with three prescriptions
Objective SDS-PAGE method was used to obtain the expression profile of erythrocyte membrane protein,which was used to search for the effect of three blood activating and stasis removing formula on the red blood cell membrane protein composition in the rat model of cold stasis.Methods 80 Wistar rats were randomly assigned to blank group,cold stasis model group and Buyang Huanwu decoction high-dose group and low-dose group,Shaofu Zhuyu decoction high-dose group and low-dose group,Danshenyin high-dose group and low-dose group with 10 rats in each group.The cold stasis model were made by ice water infusion and adrenalin hydrochloride injection for 15 days.Each drug group was given once a day.After the last time,diet is prohibited within 12 hours,but it can be drinking water.The composition of erythrocyte membrane proteins was analysed by SDS-PAGE.Results Compared with the blank group,the percentage composition of band 1 protein,band 2 protein,band 3 protein and band 7 protein were decreased significantly (p<0.05).Compared with the model group,Shaofu Zhuyu decoction could increase the percentage of band 1 protein,band 2 protein,band 3 protein,band 4.1 protein and band 5 protein significantly (p<0.05).Danshenyin decoction could increase the percentage of band 1 protein and band 7 protein significantly (p<0.05).Buyang Huanwu decoction couldn't increased the the percentage of membrane protein significantly but have some effect to increase.Conclusion Shaofu Zhuyu decoction and Danshenyin decoction could change the composition of membrane protein with different degree.
