Regulatory effect of modified Shashen Maidong Decoction on the PTEN/PI3K/AKT pathway in rats with chronic atrophic gastritis
Objective To observe the regulatory effect of modified Shashen Maidong Decoction on the apoptosis of gastric mucosa and PTEN/PI3K/AKT pathway in rats with chronic atrophic gastritis(CAG).Methods Sixty Wistar rats were randomly divided into six groups:a blank group,a model group,a positive control group,and low-,medium-,and high-dose groups of modified Shashen Maidong Decoction,with 10 rats in each group.Except for the control group,other rats drank N-methyl-N'-nitro-N-nitrosoguanidine(MNNG)solution to replicate the CAG model.The positive group was treated with vitamin C as a positive control,and 0.3 g/kg vitamin C suspension was administered by gavage after the completion of modeling.The low-,middle-,and high-dose groups were treated with different doses of modified Shashen Maidong Decoction by gavage,administered once a day for 12 weeks.The blood flow of gastric mucosa was observed by neutral red clearance.The content of Fas and FasL in serum was detected by ELISA.The apoptosis of gastric mucosa cells was detected by TUNEL staining.And the expressions of PTEN/PI3K/AKT signal pathway related proteins in gastric mucosa were detected by immunohistochemistry and Western-blot.Results After gavage treatment with different doses of modified Shashen Maidong Decoction,the blood flow of gastric mucosa was significantly increased,while the levels of Fas and FasL in serum were significantly reduced,and the TUNEL apoptosis positive cells in gastric mucosa were significantly reduced,and the number of AKT positive cells and protein expression in gastric mucosa were significantly reduced(P<0.05).The number of PTEN positive cells and protein expression in gastric mucosa were significantly increased,while the number of PI3K positive cells and protein expression were significantly decreased(P<0.05),and the effect was dose-dependent.Conclusion Modified Shashen Maidong Decoction has a good therapeutic effect on CAG rats,which can protect mucosal epithelium by inhibiting excessive cell apoptosis,and effectively inhibit the abnormal activation of PI3K/AKT signaling pathway by increasing PTEN expression,exerting a preventive and therapeutic effect on CAG.
