Effects of Jianpi Huayu Jiedu method on the expressions of AKT,mTOR and p-4EBP1 in cancer tissues and adjacent tissues of nude mouse colon cancer model
Objective To study the effects of the Jianpi Huayu Jiedu method on the expressions of protein kinase B(AKT),mammalian target protein of rapamycin(mTOR)and phosphorylated eukaryotic initiation factor 4E binding protein 1(p-4EBP1)in orthotopic implantation tumor and adjacent tissues of colon cancer.Methods A nude mouse colon cancer model was established and randomly divided into a blank group,a model group,a Jianpi Yiqi group,a Huayu Jiedu group,a Jianpi Huayu Jiedu group,a rapamycin group and a 5-fluorouracil group,with 5 mice in each group.After 4 weeks of intervention,the expression levels of AKT,mTOR and p-4EBP1 in cancer tissues and adjacent tissues were detected.Results Compared with the blank group,the expressions of AKT,mTOR and p-4EBP1 in the model group were all up-regulated,with a significant difference(P<0.05),and the expression levels of AKT,mTOR and p-4EBP1 were higher in cancer tissues than those in adjacent tissues,which in turn were higher than those in normal intestinal mucosa tissues.Compared with the model group,the Jianpi Huayu Jiedu method inhibited the expression levels of AKT,mTOR and p-4EBP1 in cancer tissues and adjacent tissues of colon cancer to a certain extent,with a statistically significant difference(P<0.05).Both the methods of Jianpi Yiqi and Huayu Jiedu down-regulated the AKT,mTOR and p-4EBP1 proteins in cancer tissues and adjacent tissues,but their reduction levels were not as high as that of the Jianpi Huayu Jiedu method.Conclusion The Jianpi Huayu Jiedu method can reduce the expressions of AKT,mTOR and p-4EBP1 proteins,and the methods of Jianpi Yiqi and Huayu Jiedu synergize it to obtain the best curative effect.
