Exploration of the mechanism of Qinggan Jiangzhuo Formula in the treatment of non-alcoholic fatty liver disease based on the abnormal expression of ERS signaling proteins
Objective To investigate the mechanism of Qinggan Jiangzhuo Formula in the treatment of non-alcoholic fatty liver disease(NAFLD)based on the abnormal expression of ERS signaling proteins.Methods A rat model of nonalcoholic fatty liver disease was induced by high-fat diet feeding.After 8 weeks of drug administration,the liver function indicators AST,ALT,AKP,the levels of serum TC,TG,NEFA,the levels of TC and TG in the liver tissue homogenate,the levels of SOD and MDA in the liver tissue homogenate were detected,and the expression levels of signaling pathway related proteins ATF4,eIF2a,and CHOP were detected as well.Results The activities of AKP,AST,and ALT in the serum,the expressions of ATF4 and CHOP,and the contents of TG in the liver tissue of the animals treated with Qinggan Jiangzhuo Formula at 865.1 mg/kg,432.6 mg/kg,and 216.3 mg/kg were significantly lower than those in the model group,whereas the activity of SOD in the liver tissue was significantly higher(P<0.01 or P<0.05,respectively).The levels of serum TC,TG,and NEFA,as well as the levels of TC,TG,MDA,and the eIF2a expression level in the liver tissue of animals in the two treatment groups of 865.1 mg/kg and 432.6 mg/kg were lower than those in the model group(P<0.01 or P<0.05).Conclusion The mechanism of Qinggan Jiangzhuo Formula in the treatment of NAFLD may be through regulating the abnormal expression of ERS signaling proteins,thereby achieving the prevention and treatment of nonalcoholic fatty liver disease.
