Electro-acupuncture pretreatment alleviating myocardial injury in ischemia-reperfusion rats by regulating AKT/mTOR pathway
Objective To observe the effect of electro-acupuncture pretreatment on myocardial ischemia-reperfusion injury(MIRI)in model rats and explore its mechanism.Methods Six SD rats were randomly selected from the 30 SD rats for a sham operation group(only threading without ligature),while an MIRI rat model was established by coronary ligation in the remaining rats,and the electro-acupuncture pretreatment,or rapamycin pretreatment,or rapamycin+electro-acupuncture pretreatment were performed on the remaining rats one week before modeling.HE staining was used to detect the pathological changes of cardiomyocytes after the modeling.The contents of CK-MB and IL-6 in serum were detected by ELISA.The protein expressions of Beclin1,Caspase3 and phosphorylated AKT and mTOR were detected by Western blot.Results Compared with the model group,the myocardial fiber breakage and interstitial bleeding were decreased in the electro-acupuncture group,and its damage degree was similar to that in the sham operation group.Compared with the rapamycin group,the myocardial fiber breakage and the degree of cell edema were reduced in the rapamycin+electro-acupuncture group.Compared with the sham operation group,the expressions of CK-MB,IL-6,Beclin1 and Caspase3 in the model group were significantly increased,while the expressions of p-AKT and p-mTOR were significantly down-regulated(P<0.01).Compared with the model group,the expressions of CK-MB,IL-6,Beclin1 and Caspase3 in the electro-acupuncture group were significantly down-regulated,while the expressions of p-AKT and p-mTOR were significantly increased(P<0.01).Compared with the electro-acupuncture group,the expressions of IL-6,CK-MB,Beclin1 and Caspase3 in the rapamycin+electro-acupuncture group were significantly increased(P<0.05 or P<0.01),while the expressions of p-AKT and p-mTOR were significantly down-regulated(P<0.01).Conclusion The electro-acupuncture pretreatment at Neiguan point can alleviate MIRI in rats,which is supposed to be related to the activation of the AKT/mTOR signaling pathway.
