Effects of Jinzhen Oral Liquid on airway inflammation and cough in model guinea pigs with bronchitis
Objective To explore the effects of Jinzhen Oral Liquid(JZOL)on airway inflammations and bronchitis cough in model guinea pigs with bronchitis. Methods Forty guinea pigs were randomly divided into 5 groups, including a model group,a blank control group,a JZOL high-dose group,a JZOL low-dose group and an amoxicillin group.A bronchitis cough model was established in the guinea pigs. After intervention, the cough sensitivity was observed in each group, the levels of serum IL-1β and CRP were detected by ELISA,the mRNA expression levels of CysLTs,MCP-1,TRPA1,CGRP in tracheal tissues were measured by qPCR,and the pathological changes in lung tissues were observed by HE staining. Results Compared with the model group,JZOL lowered the cough sensitivity in the treatment groups.Although there was no significant difference in the levels of serum IL-1 β and CRP,JZOL showed a significant tendency in reducing their levels, among which the levels of serum IL-1 β and CRP were the lowest in the JZOL high-dose group, indicating a more significant effect than amoxicillin.JZOL inhibited the mRNA expression of MCP-1,CGRP,CysLTs and TRPA1 in tracheal tissues. In addition,JZOL improved the pathological changes of lung tissues, and the degree of lesions was ranked from high to low in the JZOL low-dose group, amoxicillin group, and JZOL high-dose group. Conclusion JZOL can inhibit airway inflammation, relieve bronchitis cough, and its mechanism on relieving cough is maybe associated with regulating the TRPA1 pathway.
