单用西红花或牛黄对CCL4诱导大鼠肝纤维化程度的影响

目的 观察单用西红花或牛黄对CCL4 诱导大鼠肝纤维化程度的影响.方法 SD雄性大鼠 50只,分为空白组、模型组、阳性药组(鳖甲软肝片)、牛黄组、西红花组,每组10只.除空白组外,其余大鼠均建立CCL4 肝纤维化模型.造模1周后给药,4周后取材.测定肝、脾指数,检测血清丙氨酸氨基转移酶(ALT)、天门冬氨酸氨基转移酶(AST)、总胆红素(TBil)含量;HE染色、MASSON染色和透射电镜观察肝组织形态学改变;ELISA法检测肝脏中肿瘤坏死因子-α(TNF-α)、肝脏中白细胞介素-1β(IL-1β)和肝脏中基质金属蛋白酶抑制因子(TIMP-1)的含量;比色法检测肝组织中谷胱甘肽过氧化物酶(GSH-Px)活力程度和肝组织羟脯氨酸(HYP)含量.结果 与空白组比较,模型组肝脾指数均升高(P<0.001);血清AST、ALT、TBil含量升高(P<0.05,P<0.01,P<0.001);肝脏中HYP、TIMP-1、TNF-α、IL-1β、MDA含量升高(P<0.05或P<0.001),肝脏中GSH-Px活力和SOD活性显著下降(P<0.001).与模型组比较,牛黄组、西红花组肝脏指数及脾脏指数降低(P<0.05,P<0.01,P<0.001);血清AST、ALT及TBil含量明显下降(P<0.05,P<0.001,P<0.001);肝组织中HYP、TIMP-1、TNF-α、IL-1β、MDA含量明显下降(P<0.05,P<0.01,P<0.001),GSH-Px活力和SOD活性明显升高(P<0.05,P<0.01).结论 西红花和牛黄均能减轻 CCL4诱导的大鼠肝纤维化程度且改善肝脏功能,其机制可能与缓解炎症反应、抑制胶原生成、增强抗氧化能力有关.
Effects of saffron or bezoar alone on the degree of liver fibrosis induced by CCL4 in rats
Objective To observe the effects of saffron or bezoar alone on the degree of liver fibrosis induced by CCL4 in a rat model.Methods Fifty SD male rats were divided into a blank group,a model group,a positive drug group(Biejia Ruangan Tablets),as well as a bezoar group and a saffron group,with 10 rats in each group.Except for the blank group,CCL4 liver fibrosis models were established in all other rat groups.After one week of modeling,drugs were administered respectively,and samples were collected after 4 weeks.The liver and spleen indices were measured,and the levels of serum alanine aminotransferase(ALT),aspartate aminotransferase(AST),and total bilirubin(TBil)were detected.HE staining,MASSON staining,and transmission electron microscopy were used to observe the morphological changes in liver tissue.The ELISA method was used to detect the levels of tumor necrosis factor-α(TNF-α),interleukin-1β(IL-1β),and tissue inhibitor of matrix metalloproteinase(TIMP-1)in the liver.The activity of glutathione peroxidase(GSH-Px)and the content of hydroxyproline(HYP)in liver tissue were detected by colorimetry.Results Compared with the blank group,the liver and spleen indices were all significantly increased in the model groups(P<0.001),the levels of AST,ALT,and TBil in serum were all increased(P<0.05,P<0.001,P<0.001),and the contents of HYP,TIMP-1,TNF-α,IL-1β,and MDA in the liver were all increased(P<0.05 or P<0.001),while the activity of GSH-Px and SOD in the liver was significantly decreased(P<0.001).Compared with the model group,the liver index and spleen index were decreased in the bezoar group and the saffron group(P<0.01,P<0.001),the levels of serum AST,ALT,and TBil were significantly decreased(P<0.05,P<0.001,P<0.001),and the contents of HYP,TIMP-1,TNF-α,IL-1β,and MDA in liver tissue were all decreased significantly(P<0.05,P<0.01,P<0.001),while the activity of GSH-Px and SOD improved significantly(P<0.01,P<0.001).Conclusion Both saffron and bezoar can alleviate the degree of CCL4 induced liver fibrosis in rats and improve liver function,whose mechanism may be related to alleviating inflammation,inhibiting collagen production,and enhancing antioxidant capacity.
