Effects of saffron or bezoar alone on the degree of liver fibrosis induced by CCL4 in rats
Objective To observe the effects of saffron or bezoar alone on the degree of liver fibrosis induced by CCL4 in a rat model.Methods Fifty SD male rats were divided into a blank group,a model group,a positive drug group(Biejia Ruangan Tablets),as well as a bezoar group and a saffron group,with 10 rats in each group.Except for the blank group,CCL4 liver fibrosis models were established in all other rat groups.After one week of modeling,drugs were administered respectively,and samples were collected after 4 weeks.The liver and spleen indices were measured,and the levels of serum alanine aminotransferase(ALT),aspartate aminotransferase(AST),and total bilirubin(TBil)were detected.HE staining,MASSON staining,and transmission electron microscopy were used to observe the morphological changes in liver tissue.The ELISA method was used to detect the levels of tumor necrosis factor-α(TNF-α),interleukin-1β(IL-1β),and tissue inhibitor of matrix metalloproteinase(TIMP-1)in the liver.The activity of glutathione peroxidase(GSH-Px)and the content of hydroxyproline(HYP)in liver tissue were detected by colorimetry.Results Compared with the blank group,the liver and spleen indices were all significantly increased in the model groups(P<0.001),the levels of AST,ALT,and TBil in serum were all increased(P<0.05,P<0.001,P<0.001),and the contents of HYP,TIMP-1,TNF-α,IL-1β,and MDA in the liver were all increased(P<0.05 or P<0.001),while the activity of GSH-Px and SOD in the liver was significantly decreased(P<0.001).Compared with the model group,the liver index and spleen index were decreased in the bezoar group and the saffron group(P<0.01,P<0.001),the levels of serum AST,ALT,and TBil were significantly decreased(P<0.05,P<0.001,P<0.001),and the contents of HYP,TIMP-1,TNF-α,IL-1β,and MDA in liver tissue were all decreased significantly(P<0.05,P<0.01,P<0.001),while the activity of GSH-Px and SOD improved significantly(P<0.01,P<0.001).Conclusion Both saffron and bezoar can alleviate the degree of CCL4 induced liver fibrosis in rats and improve liver function,whose mechanism may be related to alleviating inflammation,inhibiting collagen production,and enhancing antioxidant capacity.
