Effects of Wenzhen Yunqi formula on cognitive function in rats with chronic fatigue syndrome
Objective To investigate the effects of Wenzhen Yunqi formula(WZYQ formula)on cognitive function in rats with chronic fatigue syndrome(CFS)and its possible mechanism of action.Methods Forty rats were divided into 10 each in the blank group,CFS model group,WZYQ formula group,and donepezil hydrochloride group,and a CFS model was established using the multiple chronic stress method in rats of all the groups except the blank group.After 30 days of modeling,1.039g/(kg·d)of Wenzhen Yunqi formula was given by gavage in the WZYQ formula group,and 0.51mg/(kg·d)of donepezil hydrochloride was given by gavage in the western medicine group,while 10ml/kg of saline was given by gavage in the blank group and the model group,daily for 4 continuous weeks.After that,behavioral tests were performed in the four groups.The levels of DA,ChAT,and GABA were detected by the enzyme-linked immunosorbent assay(ELISA)and the levels of pentraxin(5-HT)and neurogranulin(Ng)were detected by immunohistochemical staining after the behavioral tests.Results Compared with the model group,the positioning navigation latency,number of platform jumping error,and central compartment dwell time were decreased in both the Chinese medicine and western medicine groups(P<0.01 or P<0.05),while the platform jumping latency,number of platform crossing,platform quadrant swimming time,exhaustion swimming time,and number of hanging tail struggle were all increased in both the Chinese medicine and western medicine groups(P<0.01 or P<0.05),and the horizontal movement time,vertical movement time and frequency were increased in the Chinese medicine group compared with the model group(P<0.01),while the hanging tail rest time was decreased(P<0.01).The expression levels of DA,5-HT,GABA,ChAT and Ng were all increased in both the Chinese medicine group and the western medicine group(P<0.01 or P<0.05).Conclusion Wenzhen Yunqi formula can regulate the expression of central neurotransmitters,ChAT,and Ng,reduce somatic fatigue,alleviate negative emotions,and improve cognitive dysfunction in CFS rats.
